Age-related Changes in Bone Architecture Vary among Inbred Strains of Mice

چکیده

INTRODUCTION: There is strong evidence for genetic contributions to bone mass and architecture in mice and humans. Inbred mouse strains have recently been established as a valuable tool for studying the genetic regulation of skeletal phenotypes. We previously reported sex-related differences in age-related bone loss in C57Bl/6J mice [1] and mouse strain-related differences in the skeletal response to OVX [2]. However, there are few data showing age-related changes in bone architecture in different mouse strains. A better understanding of age-related changes in trabecular and cortical bone will enhance interpretation of murine studies with therapeutic and/or genetic interventions. In addition, characterizing the effect of age on skeletal morphology in different mouse strains may improve understanding of the genetic mechanisms underlying skeletal fragility in humans. Therefore, the objective of the current study was to assess age-related changes in trabecular and cortical bone in different inbred mouse strains.

Karyotype of NIH, C57BL/6 and Razi strains of laboratory mice (Mus musculus)

The laboratory mouse is recognized as the pre-eminent model for genetic research. Awareness of chromosomal patterns of experimental animals increases their value for a variety of different fields of study. We aimed to study mitotic chromosome preparations from NIH, C57BL/6 and Razi strains of mice, which are outbred, inbred and partially inbred laboratory mice respectively. Bone marrow cells we...

Computational Model Predicts Genetic Differences in Bone Tissue Age and Mineralization

Introduction. Genetic factors are generally acknowledged to contribute to skeletal fragility and osteoporotic fracture risk. However, the functional links that characterize how genotype affects phenotype (fracture risk) are not well understood. Previously we demonstrated that differences in femoral stiffness, strength, and brittleness among inbred mouse strains (e.g, A/J, C57BL/6J, C3H/HeJ) at ...

Inbred Strain-Specific Response to Biglycan Deficiency in the Cortical Bone of C57BL6/129 and C3H/He Mice

Inbred strain-specific differences in mice exist in bone cross-sectional geometry, mechanical properties, and indices of bone formation. Inbred strain-specific responses to external stimuli also exist, but the role of background strain in response to genetic deletion is not fully understood. Biglycan (bgn) deficiency impacts bone through negative regulation of osteoblasts, resulting in extracel...

Variation in senescent-dependent lung changes in inbred mouse strains.

Previous studies from our laboratories showed lung development differences between inbred strains of mice. In the present study, the C57BL/6J (B6) and DBA/2J (D2) strains were examined for senescent-dependent differences with respect to the lung structure and function. Specifically, we hypothesize that senescent changes in lung vary between strains due to identifiable gene expression difference...

Differences in vertebral structure and strength of inbred female mouse strains.

This study assessed mouse strain-related differences in vertebral biomechanics and histomorphometry in inbred mice strains shown to differ in bone mineral content (BMC) and areal density (BMD) (as measured by pDEXA). Lumbar vertebrae L3 to L5 were collected from three mice strains (C3H/HeJ[C3], C57BL/6J[B6], and DBA/2J[D2], n=12/strain, 4-month-old female, 22.2 +/- 0.3g). BMC and BMD were measu...
